Twelve. That's how many people took GlyNAC in the trial that made it famous. Twelve more took a placebo. Sixteen weeks later the supplemented group had walked 19% faster, cut interleukin-6 by 83%, and improved insulin resistance by 68%.
Last issue I promised we'd find out whether a trial that small can carry a claim that big. It can't — though not for the reason most sceptics reach for. The problem isn't that GlyNAC did nothing.
It's that a small trial reporting a huge effect is telling you something about the trial, not the molecule.
That rule is worth more than everything else here, GlyNAC included — and by the end you'll apply it to any headline in about fifteen seconds. Fair warning: nothing in this issue earns a B.

🔬 Top 3 Interventions Under the Microscope
GlyNAC is sold as one thing: glutathione, refilled, ageing rolled back. Underneath are three claims with three different amounts of evidence behind them — and the sturdiest belongs to the ingredient nobody markets.
1. GlyNAC for reversing ageing hallmarks — Evidence Grade: C+
The question: do those headline numbers mean what they look like they mean?
What it is. Glutathione is your cells' main internal antioxidant, built from three amino acids — and cysteine and glycine are the two that run short with age. GlyNAC supplies both, on the theory that ageing cells aren't broken so much as under-supplied.
Human evidence. Kumar and Sekhar's 2023 randomised trial in the Journals of Gerontology: 24 older adults, half on GlyNAC, half on an alanine placebo, sixteen weeks. Gait speed up 19%, IL-6 down 83%, TNF-α down 58%, insulin resistance down 68% — changes within the supplemented group, not placebo-adjusted differences.
Now the part that decides the grade. The small trials that reach publication are disproportionately the ones that landed high — so when the implausible turns up in a sample this small, the sample is usually the explanation. I'd like to be wrong. The Deep Dive shows my working.
Cautions. Nothing alarming in the trial itself. The caution is interpretive — near enough every human GlyNAC result comes from one research group, whose institution holds the patent. That isn't an accusation of bad faith. It's a reason to wait for someone else.
Takeaway. Real randomised data, honestly reported — but single-lab, tiny, and carrying effect sizes that strain belief. C+, and one that could become a B overnight if an independent group replicates even half of it.
2. GlyNAC for correcting a glutathione deficit — Evidence Grade: C
The question: forget "ageing" — does it work in people who are genuinely short?
I wanted to give this a B. I can't, and the reason is a small lesson in itself.
The largest GlyNAC trial to date came from Nestlé Research in 2022, in Frontiers in Aging: 114 healthy older volunteers, three doses of glycine and NAC against placebo, two weeks. Primary endpoint, total glutathione. It missed — placebo finished at 903.5 mg/L, the top dose at 959.6, p = 0.278.
Then the interesting bit. In participants who started with low glutathione and high oxidative stress, glutathione did rise on the higher doses. That's the finding the category leans on, and the one I'd most like to believe. But it was post-hoc: a subgroup carved out after the fact, two doses pooled, measured as change within those people rather than against placebo.
Which hands you the second rule, and it's the one that catches most people: primary endpoint first, subgroup second. A trial's main question is declared before anyone sees the data, precisely so nobody can go hunting afterwards. When the exciting number arrives from a group carved out later, you're seeing the best view of the data rather than all of it.
Cautions. There's also no routine, affordable test for tissue glutathione status the way there is for vitamin D. Even if the deficit theory is right, you can't easily find out whether it applies to you.
Takeaway. A plausible idea with a suggestive but post-hoc signal behind it. C. Four issues running now — vitamin D, magnesium, taurine, this — benefit concentrates in deficiency and thins out in sufficiency. The pattern is holding.
3. Glycine on its own — Evidence Grade: C+
The question: what if the more interesting half is the one that costs nothing?
Strip the branding off and GlyNAC is two commodity amino acids. One has ageing data that is, by this field's standards, unusually hard to wave away.
The US National Institute on Aging runs the Interventions Testing Program precisely because single-lab animal results kept failing to replicate: every compound is tested at three independent sites, to a shared protocol. Glycine went through it. In Aging Cell in 2019, Miller and colleagues found an 8% glycine diet extended median lifespan by roughly 4% in females and 6% in males. Two sites reached conventional significance; the third landed at p = 0.053 — by the standard I've applied to everyone else, a near-miss rather than a hit.
Even so, a small effect pointing the same way across three sites beats a big effect from one. Third rule: one lab is a hypothesis, three labs is a finding. Consistency check, though — this is still mice, at a dose no human eats, and I graded taurine's mouse result C− three weeks ago. Glycine earns the notch for the replication, not the biology.
And here's the part I find quietly delightful. The stack sells on its glamorous half — the antioxidant, the patent, the press releases. The half with the best ageing data is the one nobody ever bothered to trademark. It also has real human evidence for something else: a B for sleep in Issue 08, at 3 g before bed. The only B-grade thing in this issue, and it costs about a pound a month.
Cautions. The ITP dose is enormous in human terms and nobody should reverse-engineer it. Glycine itself is about as benign as supplements get.
Takeaway. The best-replicated ageing result in this issue belongs to the cheapest ingredient in the bottle. C+ — animal-only, but animal-only of a better kind.
SPOTLIGHT
⭐ Spotlight: Should You Take an Antioxidant Every Day?
GlyNAC's whole logic is that more glutathione is better. That deserves a harder look than it gets, because two decades of antioxidant research have been an extended argument against exactly that assumption.
Pros. N-acetylcysteine isn't a fringe compound. It's a licensed medicine — the hospital antidote for paracetamol overdose — with decades of safety data behind it. And the biochemistry is sound: cysteine really is the rate-limiting ingredient for glutathione, so supplying it does raise production when supply is what's limiting.
Cons. Reactive oxygen species aren't simply damage. They're also a signal — the message telling muscle to build mitochondria and cells to switch on their own defences. Mop them up indiscriminately and you dampen that signal. In a double-blind trial in The Journal of Physiology in 2014, Paulsen's group found vitamin C and E supplementation blunted the cellular markers of mitochondrial adaptation to endurance training. Being precise: performance improved equally in both groups. What suffered was the machinery underneath. Readers of Issue 14 will recognise it — the cold plunge problem, in a capsule.
A second concern, flagged carefully. NAC has accelerated tumour progression and metastasis in mouse models of lung cancer and melanoma — Sayin's 2014 work and Le Gal's 2015 paper in Science Translational Medicine. I spent all of last issue arguing that mouse results don't automatically transfer, and honesty means that cuts both ways: mouse harm may not transfer either. But when the same molecule shows mouse benefit and mouse harm, taking the benefit seriously and shrugging off the harm isn't scepticism. It's shopping.
Bottom line: ⚠️ Promising but premature. The problem was never the molecule. It's the framing — "take an antioxidant every day, indefinitely, whether or not you're short" is a proposition nobody has tested for long enough to endorse. |
DEEP DIVE
📖 Deep Dive: How Twelve People Made a Very Big Claim
Why should a 68% improvement from twelve people reassure me less than a 5% improvement from five thousand?
It feels backwards. Bigger result, better news — that's the intuition, and it's wrong in a way that has quietly misled a generation of supplement buyers. The Deep Dive works through it in plain English, and ends with the four things GlyNAC would need to show to earn a B. There's also a detail in the Nestlé story I genuinely didn't expect to find, and it's to their credit.
HYPE CHECK
🚨 Hype Check: The Patent, Not the Discovery
The Hype: Branded GlyNAC sells as a premium category — "cellular nutrition," glutathione replenishment, ageing addressed at the cellular level. Nestlé Health Science markets it through its Solgar Cellular Nutrition line as Cellular Protect (previously Celltrient), at a substantial multiple of what the ingredients cost.
The Evidence: Follow the paperwork. Baylor College of Medicine holds a patent on the glycine-plus-NAC combination and licensed it to Nestlé Health Science. Nestlé Research then ran the largest GlyNAC trial published to date — the 114-person study above — and it missed its primary endpoint. It's the biggest human GlyNAC study in existence, and the one you'll never see quoted in an advert.
Why It's Misleading: You're being sold a patent, not a discovery. Glycine and N-acetylcysteine are two of the cheapest compounds in the trade, individually unpatentable and available in bulk for a few pounds a month. What's proprietary is the pairing — and no trial has ever compared branded GlyNAC against the same two amino acids bought separately.
Our Verdict: ❌ Not at that price. Plain glycine and NAC powders cost a fraction as much and are chemically the same thing arriving in the same order. Spend the difference on food, or on the patience it'll take for someone independent to check the headline. The molecule might yet earn its reputation. The packaging has already spent it. |
SUPERFOOD
🍲 Superfood Spotlight: Collagen-Rich Cuts
An awkward fact for the supplement aisle: the glycine half of GlyNAC is abundant in the cheapest meat in the shop.
Glycine makes up roughly a third of the amino acids in collagen, so it concentrates in the bits modern cooking discards — oxtail, shin, chicken wings and skin, anything with connective tissue. Gelatin runs to around 19 g of glycine per 100 g; a properly made bone broth carries roughly 1–2 g per cup. That won't reach the 7-odd grams a day used in the GlyNAC trials, but it comfortably covers the 3 g behind our Issue 08 sleep grade.
Two honest caveats. It gives you glycine, not cysteine — half the pair. And bone broth's other reputation, the gut-healing and joint-repair claims, isn't supported by good evidence; we graded collagen peptides F for everything but skin in Issue 13.
One useful test: real broth sets to a wobble in the fridge. If your carton stays liquid, it's stock, and there's very little collagen in it. A packet of oxtail is about £4 — and unlike everything else in this issue, it's genuinely nice to eat.
BIOHACKING CORNER
🌡 Biohacking Corner: The Cheap Half, Done Properly
Nobody has shown that GlyNAC extends a human life. But one of its ingredients is cheap, evidenced for something else, and easy to get from food — so here's what I'd do while we wait.
1. Cook the connective tissue. One slow-cooked collagen-rich meal a week, or a mug of broth that passes the fridge test — glycine as food rather than as a subscription.
2. If you want the powder, buy it for sleep. Three grams of plain glycine before bed is the dose behind the B in Issue 08 — a few pounds a month, for something you can notice within a week.
3. Don't blanket-dose antioxidants around training. If you lift or run for adaptation, keep high-dose antioxidants away from the hours after a hard session — same timing logic as the cold plunge in Issue 14.

Personal note: I've had 3 g of glycine on the bedside table since Issue 08 and it's stayed there — the one thing in this issue I'd have bought anyway. I haven't added NAC. Not because I think it's dangerous, but because "indefinitely, for a deficit I can't measure" isn't a protocol, it's a vibe. |
Caveat: most bronchospasm reports with acetylcysteine involve the inhaled form rather than oral supplements, but if you have asthma it's worth raising. NAC can also interact with nitrates. Discuss it with your doctor if you're pregnant, breastfeeding, on medication, or have a history of cancer. |

They cost nothing and they work on everything.
That's three issues running where the capsule was the least interesting thing on the table. So Issue 21 leaves the supplement shelf for muscle — protein dose in older adults, what resistance training does to mortality curves, and a Hype Check on the "muscle preservation" powders sold to people who could just lift something.
